Unleashing the Power of End-to-End Automation in Biologics Development with the Process Analytical Sciences Central Automation Lab (PASCAL): Orchestrated Hands-free Analytical Testing at Scale
Presented by: John Derent, Senior Automation Engineer, The Janssen Pharmaceutical Companies of Johnson & Johnson, Biosero
Biotherapeutics development requires rigorous analytical testing in support of process development, formulation, and product characterization to achieve optimal manufacturing of safe and effective protein, cell, and gene therapies. The throughput and scale of analysis contribute to limitations in both speed and capacity of biologics clinical development. In this talk we will share our design, build, and implementation journey of PASCAL (Process Analytical Sciences Central Automation Lab): orchestrated modular end-to-end physical and digital automation of biologics development analytics at scale. We will share the first phase of this bold industry-leading automated laboratory, showcasing hands-free experimentation and data flow to reduce experiment cycle time by 6-fold, while increasing analytical capacity 4-fold. We will share our strategy for phased expansion to over 50 workflows and goals of additional implementation to provide life-saving therapies to patients faster.
Streamlining DNA Sequencing: Automation-Friendly and Expedited Workflows with the NEBNext UltraExpress FS DNA Kit
Presented by: Siva Chavadi, Ph.D - Sr. Field Application Scientist, NGS US-EAST, New England Biolabs
The NEBNext UltraExpress®Library Prep Kits represent the latest generation of NEBNext DNA and RNA library prep workflows that are designed to enhance the efficiency of NGS sample preparation. This presentation will discuss the key features and benefits of the NEBNext UltraExpress FS DNA kit, focusing on its ease of use, cost-effectiveness, and compatibility with various automation platforms.
The kit is optimized for DNA inputs ranging from 10 to 200 ng, providing a single protocol for key workflow parameters including adaptor dilution and PCR cycling conditions across the recommended input range. This uniform approach facilitates integration with automated platforms, making it suitable for high-throughput applications.
The streamlined workflow of the UltraExpress FS DNA kit eliminates the need for a post-ligation bead cleanup and consolidates multiple enzyme reactions into a single mastermix. The result is a single-vessel, addition-only process, reducing reagent and plastic consumption, as well as associated costs. Incorporated enzyme-based fragmentation obviates the need for expensive capital equipment and consumables, while saving time and improving yields
By leveraging the automation-friendly features of the NEBNext UltraExpress FS DNA kit, laboratories can achieve consistent and reproducible results, minimize hands-on time, and increase throughput, thereby advancing research capabilities.

Embark on a transformative journey toward sustainable and energy-efficient sample storage practices. This presentation contextualizes sustainability within global and industry-specific frameworks, emphasizing the growing imperative for energy efficiency. Through compelling data and real-world challenges, we’ll illustrate why sustainability must be prioritized in sample management.
The session will spotlight Azenta’s latest innovation: a high-density -80°C automated sample storage system. This cutting-edge solution promises significant savings, cost reductions, and environmental benefits. Attendees will gain first-hand insights into the system’s capabilities and explore its potential to revolutionize sample storage –most recognised when compared to replacing a large manual ULT freezer collection with an Azenta Life Sciences automated storage and retrieval system. By modeling time, power, space, carbon emissions, and running costs, we demonstrate a 77% reduction in electricity consumption and carbon emissions, an 83% reduction in floorspace, and a 40% reduction in labor hours. The session aims to address pressing challenges and share best practices for advancing energy efficient, sustainable, and cost-effective sample and material management. Join us as we pave the way for a sustainable and energy-efficient future in sample storage.
Revolutionizing Lab Sustainability: Plasma Cleaning for Plastic Reuse
Presented by: Paul Hensley, IonField Systems CEO, IonField Systems
Atmospheric Pressure Plasma (APP) offers a breakthrough approach to the cleaning and extended reuse of plastic lab consumables like pipette tips and microplates by harnessing a potent mix of oxygen radicals, hydroxyl radicals, and excited nitrogen species.
Oxidative destruction conceptually is a microscopic fire—instead of flames, the energy in reactive oxygen species tears molecules apart, dismantling contaminants at the atomic level. A process that leaves no residue. Unlike traditional solvent-based methods, plasma cleaning erases biological and organic chemical residues at the molecular level, modifying surface chemistry without structural damage.
In this talk, we will discuss the mechanisms behind plasma cleaning, its advantages over purely solvent-based decontamination, and real-world applications demonstrating how this technology extends the lifecycle of plastic consumables while maintaining performance standards.

InTraSeq™ technology: a multimodal single cell method for simultaneous measurement of RNA and Proteins to uncover new single cell biology
Presented by: Greg Warner, Ph.D - Sr. Field Applications Specialist, Cell Signaling Technology
InTraSeq™ is a novel technology for single cell analysis that enables unbiased measurement of both RNA and proteins in individual cells. This innovative approach employs validated and functionally tested antibodies at CST, which allows the identification of cell subtypes and Signaling Pathways in single cells by measuring post-translational modifications (PTMs).
One of the significant advantages of using InTraSeq™ is its ability to quantify extracellular, cytoplasmic, and nuclear proteins, including transcription factors, and their PTMs while preserving the RNA and heterogeneity of the sample. This method offers an extensive view of the cellular mechanisms of complex biology, which was previously missing in single cell RNA sequencing datasets. Researchers can use InTraSeq™ to accurately characterize single cell acute and chronic responses, uncover new cell states, and more accurately analyze cell differentiation trajectories. Moreover, InTraSeq™ offers a streamlined protocol that requires only one hour of benchwork and allows for multi-day sample storage, making it a practical and efficient tool for high-throughput applications.
