The measurement of binding affinities is an essential part of the drug discovery process. From biomarker discovery, to target validation and lead optimization, label-free assays are beneficial and critical. While SPR is broadly used, it has numerous problems especially when trying to measure small molecule binding where DMSO may be present. iRiS Kinetics is a new label-free measurement technology that is not affected by the presence of DMSO. Its flow cell design results in good kinetics measurements, free of diffusion effects as in BLI.

PHERAstar FSX with Advanced Assay Stability (AAS) System helps to increase data quality of high-throughput screening assays

Experimental results can be substantially altered by temperature which can affect many events including the efficiency of biochemical reactions. Since biochemical reactions are at the heart of 2 of the most widely used detection methods, luminescence and AlphaScreen, concern about unreliability of the outcome for these assay is prevalent. This is especially true as higher density plate formats such as 1536 well are used that necessarily have longer read times and thus are more greatly affected by the difference in reader temperature compared to laboratory temperature. With the AAS you no longer need to worry about a temperature difference between the reader and the lab environment. Furthermore, for screening campaigns that can last for multiple hours you can be sure that the first and last plate will be read under the same conditions! In addition the AAS exhibits reduced sample evaporation compared to other readers. Also the cooled air provided by the AAS first moves through the readers interior so the measurement system and excitation sources are also cooled and kept at optimal operating conditions. We are excited to be able to offer this innovative system that does not substantially increase the footprint of the PHERAstar FSX with no additional tubes or devices required for its operation.

Engineering an Automated Platform for Large-Scale Production of Plasmid DNA without In-Process Cross-Contamination

The global demand for plasmid DNA has increased rapidly since the advent of cell and gene therapies (CGT) followed by hyper-growth during the outbreak of the COVID-19 pandemic. These cutting-edge therapies come in many forms but all of them rely on plasmid DNA at some point either in their manufacture or mechanism of action. Unfortunately, scaling up manufacturing capacity for plasmid DNA is a difficult operation and it has become a bottleneck for the CGT industry. Most companies outsource plasmid production to contract development and manufacturing companies (CDMOs) who are specialists in the area and have in-house manufacturing platforms. Here we present GenScript’s new high-throughput plasmid production platform designed to produce up to 960 low-endotoxin plasmids at the 1 mg scale per 24 hours. This integrated platform brings the best of industrial automation to a biological manufacturing process. The platform contains six industrial robot-operated modules encompassing inoculation, incubation, pelleting, lysis, chromatography, and solvent extraction. It can operate continuously with in-process cleaning to prevent cross-contamination. Our platform can produce transfection-grade plasmid libraries for AAV capsid and vector screening, genome-wide CRISPR libraries, and in vitro transcription (IVT) templates for mRNA production for the CGT industry. GenScript will accelerate the commercialization of new CGT products by providing high quality plasmids with a two-day turnaround time. 

Extraction of Total Nucleic Acids from FFPE Samples using Adaptive Focused Acoustics® (AFA®) Technology

Comprehensive Genomics Profiling (CGP) enables molecular pathology laboratories consolidate individual biomarkers into Next-Generation Sequencing (NGS) assay (Dy, Nesline et al. 2019, Nesline, DePietro et al. 2019). While formalin-fixed paraffin-embedded (FFPE) methods continue to gain popularity owing to their ability to preserve human tissue specimens, they also pose several challenges – from complex paraffin removal processes to extracting adequate amounts of DNA and RNA, from expediting the turnaround time to increasing throughput. Technology improvements in liquid chromatography/mass spectrometry have enhanced the detection and identification of metabolites and lipids from complex biological samples. As metabolomics and lipidomics measurements become increasingly valued, there is a growing need to automate sample preparation workflows. Specifically, Agilent automation offers intuitive workflows that provide high data reproducibility and increased throughput while reducing hands-on time. Here, we describe key learnings revealed during the automation of several workflows that extract metabolites and/or lipids from plasma and mammalian cell samples.

Recent Advancements in Automated Liquid Handling

Automated liquid handling has become necessary in labs to cut down on manual work and keep up with growing workloads while maintaining efficiency, reproducibility, and careful use of resources. Formulatrix has introduced cutting-edge liquid handling technologies for routine lab operations like serial dilution, normalization, cherry picking, plate broadcasting, pooling, etc. In this presentation, I will introduce our two new liquid handlers, both with different pipetting technologies: The F.A.S.T., a 96-channel positive displacement liquid transfer system, and the FLO i8, a fully independent 8-channel air displacement liquid handler with automated liquid transfer optimization. The F.A.S.T. (Flow Axial Seal Tip) is a reliable and easy-to-use liquid transfer system designed to quickly move any viscosity of liquid with its one-of-a-kind positive displacement, 96-channel head. Its high accuracy, precision, and reliability for low-volume sample transfers starting from 0.1 ul saves researchers time and reduces reagent costs. The FLO i8 Liquid Handler is ideal for optimizing the protocols and simplifies experimental design, and execution. Its intuitive software, flexible hardware, and intelligent pipetting guarantee accurate and precise liquid transfers without specifying source volumes, labware geometries, and liquid properties.

Improving Efficiencies in Automated Workflow Solutions by Integrating the Thermo Scientific Multidrop Combi SMART+ Automated Reagent Dispenser

A Novel Method for High Throughput Lyobead Formation in Diagnostic Kit Manufacturing
