Novel Technology Presentation Abstracts:

Washing and Reusing Tips Using Automated Tip Washers in Production Labs

Disposable pipette tips provide an easy process for liquid transfers. Considered for one-time use, repeated pipette tip purchases incur ever increased costs and decreased profitability. In addition to cost, once discarded, these polypropylene plastics enter the waste stream and are either incinerated as biohazard waste or discarded to a landfill. Because of the lack of any high throughput cleaning technology, washing pipette tips for reuse was not generally accepted as safe for many assays.

Using a proven technology, Grenova tip washing devices demonstrate that pipette tips can be safely and effectively washed for many laboratory assays. Grenova tip washers have been validated in clinical laboratories and are being used in PCR, immunological, qPCR for NGS library generation, toxicology, LC-MS/MS, NMR, and GD-FID production assays. Furthermore, Grenova’s tip washing technology has been successfully tested for cell culture, high throughput pharmaceutical compound assay testing for compounds in DMSO, and forensic toxicology testing.

Grenova’s tip washers are brand agnostic and can be designed to work with any 384, 96, or 24 industry standard tip. Grenova also offers integration options for a fully automated workflow. Advances in Tube Handling

The design also has a positive impact on the throughput. Due to minimum distance between the sample to be picked and the target rack, the pick and place operation can be performed much faster. The Tube Handler HT700 was developed in collaboration with leading research laboratories worldwide to maximize efficiency, save benchtop space, and enhance automation.

Enabling Ultra High-Throughput Mass Spectrometry With Acoustic Mist Injection

Mass spectrometry has the unique ability to qualitatively identify and precisely quantify nearly any analyte and therefore it has become an attractive candidate to generate label-free screening data. However, existing sampling methods do not reach the throughput and assay volume standards to justify including mass spec in high-throughput screening. While MALDI-TOF mass spec meets these requirements, the matrix used in the experiment often interferes with the small molecule analytes in the reaction. Given the limits of existing methods and Labcyte’s groundbreaking acoustic liquid handling, we set out to determine whether our technology could be applied to mass spec systems.

Labcyte’s Acoustic Droplet Ejection technology has been used for over a decade for millions of tipless transfers to help minimize reaction volumes to nanoliter scales. Here we show acoustic technology can create a femtoliter mist that can be ionized and directly injected into a mass spectrometer, resulting in sensitive and reproducible mass spec data. In addition to this new loading technology, the system samples at a rate much faster than existing methods, three injections per second. Given these parameters, this system can generate ultra high-throughput mass spec screening data of a million-compound screen in approximately two work weeks. To demonstrate the power of this method, we present an acoustic system that can inject sample directly from the assay well into a mass spectrometer which is then used to monitor the quantitative ratio of product to substrate. This experiment, developed in only two weeks, culminated in a 240,000-compound screen completed in 66 hours on a prototype system. We believe Acoustic Mist Injection has the potential to impact label-free high-throughput screening in the same way Acoustic Droplet Ejection revolutionized liquid handling a decade ago.

Optimizing Automated Liquid Handling Protocols with the BRAND LHS Software

Designing a liquid handling protocol for automation for the most efficient use of time and materials can be tedious, with a lot of trial and error. BRAND’s new, compact Liquid Handling Station (LHS) features extremely user-friendly software that not only makes initial protocol design easy, but simplifies the optimization process. Easy drag-and-drop rearrangement of the instrument deck and a 3D simulation model allows quick determination of the best layout and process verification. Agilent Automation – AssayMap Bravo – Automated Sample Prep for Proteins and Peptides

The AssayMap Bravo provides a unique technology for protein and peptide sample prep. This automated platform has workflows established for affinity purification, in-solution digestion, protein and peptide clean-up (desalting), fractionation, phosphopeptide enrichment and N-Glycan analysis currently. Using the AssayMap Bravo platform allows for samples (1-96 wells) to be prepared with reduced variability, provides for a clear path for sample processing scalability and enables customized workflow development without the need for in-house automation expertise.

Hidex Sense Multi-Mode Plate Reader

The Hidex Sense is a compact, state of the art, multi-technology microplate reader providing a powerful, yet flexible platform for performing any kind of assay without limitations. The amazingly compact design, makes it ideal for even the smallest laboratory, and allows the unit to be easily adapted for robotic integration or placement within environmental control chambers. The system software is extremely user friendly – rated ‘Excellent’ on the System Usability Scale (SUS) – and as such, requires minimal training.  The software includes a comprehensive library and a variety of data analysis tools, including export of data files into Excel. The uniquely designed Spectrograph for absorbance detection, allows reading of full spectrum in less than a second per well, with ratio analysis possible in just one reading. Temperature control of up to 65C, as well as linear, orbital and double orbital shaking are standard on the system, as well as optional dual-dispensers and atmospheric control.

The Sense Beta Plus version of the reader uniquely combines liquid scintillation counting with all common non-radiometric measurements, such as absorbance and fluorescence, into one very compact unit.

The dedicated PMT detector for LSC is also used to obtain ultra-high sensitivity luminescence for demanding applications and for reducing reagent use in assays such as dual luciferase, providing up to ten times more sensitivity than other common plate readers.
