How automation helped increase workflow efficiency during the peak of COVID testing

When the COVID-19 pandemic started, accurate testing became a top priority for countries around the world. As testing ramped up, labs suddenly faced an exponential increase to their usual number of samples to process. Unfortunately, many of these labs were lagging due to manually intensive practices. Over the course of the pandemic, however, the testing response time improved tremendously thanks to the power of automation.  

Hamilton Storage’s line of cappers and decappers allows labs to increase throughput while reducing the risk of cross-contamination. Manually capping and recapping tubes is time-intensive and has the potential to create a bottleneck in the lab because there are physical limits to how many samples can be processed. It can take a person more than 10 minutes to decap 96 tubes, while it takes the LabElite DeCapper less than 60 seconds. By automating the entire workflow, cross-contamination is virtually eliminated. Naturally, this leads to increased sample safety and fewer false positives. 

As of May 2, 2022, the United States had performed nearly 1 billion tests for COVID-19–the highest number of any country worldwide. Thanks in large part to automated benchtop devices such as the LabElite DeCapper, an overwhelming majority of these tests were deemed reliable and the results helped curb further spread of the virus.

Novel Sample Collection Technologies iSWAB-MB-EL and NextSWAB Increase the Efficiency of SARS-CoV-2 Testing by Bypassing RNA Extraction Steps

Viral testing via qPCR is one of the most effective tools in pandemic control. The onset of the COVID-19 Pandemic and emergence of the Delta variant created supply chain pressures and labor shortages that contributed to major lab backlogs, compromising sample integrity and the ability to conduct large-scale testing necessary for effective pandemic responses. To combat these challenges, Mawi DNA Technologies released two novel sample collection technologies: iSWAB®-MB-EL and the NextSWAB. iSWAB®-MB-EL is an Extraction-Less sample collection solution designed to eliminate RNA extraction from COVID-19 molecular testing workflows, allowing researchers to perform direct RT-PCR on COVID samples. Bypassing time-sensitive and delicate RNA extraction steps allows labs to deliver results sooner, increase testing throughput, and reduce costs. iSWAB®-MB-EL has been validated for use with several COVID-19 EUA Molecular diagnostic tests, including traditional RT-qPCR and rapid detection RT-LAMP based methods. The iSWAB®-MB-EL solution inactivates COVID-19 and stabilizes samples at ambient temperature for at least 28 days, eliminating the need for expensive cold-chain transport. In response to swab shortages, Mawi also released the NextSWAB, a sterile, 100% injection-molded plastic applicator designed for the high-efficiency collection and release of nasal samples. The flexible head is designed with two channels separated by a septum to retain the collected nasal material until the user releases it into transport media with a gentle shake. Eliminating absorbent material allows for effective release and concentration of the specimen. The NextSWAB is a comfortable, high-performing traditional swab alternative that offers multifunctional use for sample collection without the possibility of contamination from glue or fibers used in flocked and cotton swabs. All Mawi products including iSWAB-MB-EL and NextSWAB are manufactured in the US to reduce supply chain risks.

Scaling Beyond COVID Rapid Testing

COVID is still a testing challenge for citizens, businesses, and entire countries. The pandemic afforded us (the healthcare community) the opportunity to improve processes, and most importantly to advance science. The regulatory process has streamlined approvals for testing, both in the tests used and in the labs that can perform the tests, but due the magnitude of testing, simplification and economics in testing became key drivers, and this is where rapid test technology shines. Point-of-Care (POC) Over the Counter (OTC) tests are now approved for COVID and there will be more to follow, likely in other infectious disease areas. Beyond COVID, we see a medicine cabinet filled with home tests for Influenza, Streptococcus, Pneumonia, all connected via mobile devices to your physician via telemedicine. If results are negative, only then do you proceed to Urgent Care. The applications reach far beyond infectious disease - think about food allergies. How many of us have nut, dairy, gluten or seafood allergies? When venturing out to a restaurant and verifying with the server that the food is allergy-free, would we not want to have our own portable rapid test in hand to verify that it actually is a gluten-free dish before we indulge? The point here is COVID was the door opener to simplified, affordable testing which puts the testing device in the hands of the patient and consumer. The next step is to consider how to develop these rapid tests and how these tests can be produced at scale. That is where BioDot comes in. BioDot made its mark in this industry in 1994 with patented, non-contact, nanoliter (nL) dispensing technology and then designed instrumentation to precisely dispense volumes onto a variety of surfaces. A rapid test is typically either a lateral flow test or a biochip/biosensor device. The surfaces used in lateral flow are porous and dispensing is best done without touching (disrupting) the material (non-contact). Conversely, in biochip/biosensor applications, the surfaces are often rigid plastic wells, but they require absolute placement of many different low-volumes of liquids (>36 different types) to be dispensed simultaneously and quickly. Both examples stated (lateral flow and biochip/biosensor) benefit from BioDot’s non-contact, nanoliter dispensing technology. As with most science, development of a rapid test begins with research, progresses to prototyping, and ultimately advances to production volumes once a test has been proven and is ready for sale. BioDot has a product offering to suit the needs of our customers with entry-level R&D platforms that scale as their development and production needs get defined. Our R&D systems are used to produce batches of up to 500,000 rapid tests per year. As our customer’s tests gain market adoption, with volumes greater than 1M tests per year, migrating to our high production instrumentation for printing and lamination is common. If your group has ELISA/antigen/antibody expertise and are considering a rapid test development effort, this presentation will further expand on the trends in this space and make recommendations of how to avoid pitfalls in your “rapid test” quest.

Using pneumatic technology to build high performance liquid dispensing systems. One of the central topics in laboratory automation is the handling of the smallest amounts of liquid – from milliliters to nanoliters. This requires not only absolute precision but also maximum flexibility, because the most diverse media and changing fluid properties place special demands on the solutions used. Pressure-controlled liquid dosing systems are a simple and efficient way to achieve precision, scalable liquid handling. This presentation conveys the constructive basics of such dispensing systems – including the control of flow, pressure, and other variables to optimize the throughput. In addition, selected systems, and products from Festo, which are presented in these solutions are used.

Improve Your Assay Automation Workflow with New QualAssure™ Solutions from Artel

Different liquids have different properties, and depending on their viscosity, density, volatility, etc., that can introduce challenges with your liquid handler delivery volumes and ultimately with your assay. Parameters can be adjusted in your automated liquid handler to ensure the accuracy and precision of the volumes being dispensed, but DIY methods are time-consuming. Therefore, Artel has added two additional solutions to the existing QualAssure portfolio so you don’t have to operate blindly and waste expensive assay reagents. As assay throughput increases, the limit of detection decreases, and assay volumes decrease there is a general higher level of scrutiny on quality. Automation addresses many of these issues and is becoming more prevalent than ever before. However, developing liquid classes can be a complex and arduous process. PCR master mix and serum are two common non-ideal solutions used in a variety of essential assays. Due to the unique rheological properties of these solutions, compounded by the absolute need for accurate and precise results of these assays, scientists depend on liquid handling precision and accuracy. In many cases, scientists optimize their liquid class settings with the actual reagents or biological solutions, however, this is problematic due to prohibitive cost, rarity, or resource requirements.  Simply using water or a glycerol mixture to mimic these solutions is not a suitable option since there are additional liquid class parameters that need to be taken into consideration. It is possible to reliably measure and adjust for master mix like liquids as well as various serums by using the new PCRMix™ and SerumSub™ QualAssure solutions. These solutions have liquid handling properties analogous to commonly used PCR master mixes and sera, respectively, and are used to help improve automated assay workflows.

The AssayMAP Bravo is a flexible yet easy-to-use automated protein sample preparation platform. The system leverages microchromatography and preoptimized applications to perform a wide variety of protein sample preparation workflows required during biotherapeutic development. We have made several advances that allow the AssayMAP Bravo to address even more of the drug development process. The system can now use large capacity packed resin bed cartridges allowing for more sample to be processed for downstream analysis as well as quicker time to results while maintaining reproducibility and low elution volumes.

How to adopt an Automation-First Mentality

Successful laboratory automation requires us to think in the language of automation rather than thinking about traditional workflows or related terms first (i.e., increasing throughput using the current methodology), which invariably add project risk when automation principles are applied afterward. For example, many scientists are introduced to automation through automated liquid handling devices. However, liquid handlers are highly specialized themselves, and adding multiple other specialized devices to a liquid handling deck ensures that the deck’s size will physically constrain both the amount and extent of the hands-free work that can be performed on it. More importantly, this approach to laboratory automation may limit the very range of possible solutions that we can conceive. Simply put, how we allow ourselves to think about how we conduct science determines how we are able to think about science itself. For laboratory automation to keep pace with science, concepts such as that of the modular workflow must be taken “off deck”. This allow new paradigms to emerge – ones that incorporate laboratory devices, samples distribution and software to reimagine total workflows and data use. Only in this way can laboratory automation truly adapt and re-adapt to inevitable changes in one’s science, technology, or organization. In this talk, we explore an “automation-first” journey into laboratory automation to expand the capabilities of hands-free science.
