Laser marking is rapidly becoming the gold standard in sample traceability as powerful lasers are now small enough to operate in a laboratory setting. Laser marking has several advantages compared to traditional ink and label systems and is particularly well suited for labeling cryovials for long term cold storage. The ability to quickly and irreversibly inscribe information into plastic labware is improving sample traceability in laboratories across the globe. AFYS3G has recently developed both a manual and automated laser marking system designed to fit the needs of different laboratories. This presentation will explain the process and benefits of laser marking labware and discuss the features of the newly released AFYS3G Lambda8 and Lambda576 laser markers.

Centrifugal BlueWasher: Better data from cell-based assays and tip-less, high speed separation for mag bead assays

Frank Feist, co-founder and CEO of BlueCatBio will present an overview of the centrifugal BlueWasher’s design and function, and review client publications that focus on BlueWasher assay design and results. With an installed base of > 160 BlueWashers in 16 countries since launch in 2014, the BlueWasher has rapidly become the new standard for contact-free media change and cell wash for cellular assays. With the addition of a suite of magnetic carriers, BlueWashers now also save tips and speed up magnetic bead workflows ranging from 3D cell screens to Luminex, DNA/ RNA extraction, NGS workflows such as SureSelect, AMPure and NGS library prep. For cell-based assays, the key benefit of the centrifugal BlueWasher vs. conventional, tip-based washers is the improved assay data quality. Users typically see z’ improvements in the 0.1 to 0.3 range so chemists get more reliable compound profiles - screens generate fewer false positives and false negatives. The presentation reviews publications by AZ, Merck, SCRIPPS and Merck KgaA. For mag bead based workflows, the BlueWasher eliminates tips for separation steps and dramatically increases throughput, reducing CAPX need for ALHs. The talk will review our new application note showing how the BlueWasher reduced tip cost and accelerated AmPure workflow at UCSD.
The OrganoPlate® - a high throughput microfluidic culture platform for 3D cell culture

The transition from 2D to 3D cell culture is a first step towards more physiologically relevant in vitro models for human (disease) modeling. To adequately capture the complex tissue architectures observed in vivo, 3D microfluidic techniques incorporate and achieve long-term gradient stability, continuous perfusion and patterning of cell layers as stratified co-cultures. Mimetas develops Organ-on-a-Chip-based models for disease modeling and evaluation of new medicines. Our unique microfluidic technology – the OrganoPlate® – enables testing of compounds on miniaturized 3D organ models in high-throughput. These models are expected to show better physiological relevance versus conventional 2D cell culture models, without compromising throughput or ease of use. This presentation provides an overview of the technology and how it can be integrated into automated workflows.

Fully-Automated 384-Well Bioassays using the Echo® Acoustic Liquid Handler

For nearly two decades Labcyte Echo® Liquid Handlers have been used in drug discovery to accurately and precisely transfer small molecule compounds, in nanoliter increments in a non-contact, tipless manner. Labcyte has advanced acoustic liquid handling by incorporating a unique real-time transfer technology called “Dynamic Fluid Analysis” to enable reliable dispensing across a wider array of fluids. Further to this, the Echo Liquid Handlers’ ability to dispense from any-well in a source plate to any-well in a destination plate provides flexibility when optimizing bioassay design, especially to eliminate location biases. Additionally, Echo Liquid Handlers for Regulatory Environments enable FDA 21CFR Part 11 compliance, locking down and tracking all Echo Liquid Handler usage, protocol changes, and file outputs via state-of-the-art security software. Partnering with experts in bioassays, Labcyte has developed methods allowing users to easily set up complex potency assays by transferring fluids directly to all plate wells, instead of sequentially diluting across the plate, resulting in less reagent usage and higher throughput while maintaining the accuracy and precision that is essential to distinguish amongst relative potencies. Here we present the Echo system as a solution to miniaturize bioassay workflows while maintaining data quality. Specifically, we present an overview of the development of two distinct bioassays: a TNF-alpha inhibition assay and the ADCC Reporter Bioassay. Through assay development, we took these two assays from manual 96-well assays to fully-automated 384-well assays. From these data and others like them, we believe the Echo system will have great impact on the field by enabling the automation and miniaturization of bioassays and quality control of biologic therapeutics.

Green Button Go automation scheduling software supports effective error recovery in real time so that samples are fully processed, even with an irrecoverable hardware error! It does this by allowing for replacement or calibration of an instrument without having to abort a run. We will present two new modules for Green Button Go - GBG Batch Planner and a liquid handler driver. GBG Batch Planner plug-in builds a calendar avoiding conflict for availability of readers and imagers. It allows interleaving of batches by multiple users to conduct kinetic studies with time-course imaging. Customers with complex scenarios of scheduling a liquid handler platform in the automated workflow can now easily integrate with a novel liquid handler driver from Biosero.

NGS Workflow: Overcoming Barriers for Successful Automation

Many labs turn to automation to increase throughput, lower costs, and reduce operator-related errors. Transferring a manual, bench-top assay to an automated platform, however, is often more complicated than expected. By understanding the key steps in complicated workflows such as NGS and taking steps to minimize variability upfront, successful assay automation can be facilitated which can save time. Examples of key steps in the process include DNA normalization, which requires a unique volume of diluent and DNA (both typically <10 µL) for each sample and addition of a master mix solution, which is typically viscous. We will highlight our approach to improving NGS workflows and present our methodologies for improving DNA normalization, ensuring accuracy of master mix addition, studying mixing efficiency and improving operator technique.
